The actor, best known for his role in television show Supernatural, shared the news during an appearance on the Live! With Kelly show earlier this week.
Padalecki told the host: "I'm a father of two. Now about to be a father of three in March."
The 34-year-old and his wife, Genevieve, started dating after meeting on the set of Supernatural in 2008. They married in Sun Valley, Idaho, in February 2010. The couple are already parents to sons Tom, four, and two-year-old Shepherd.
