An insider is quoted by the Mail on Sunday as saying the relationship "simply run its course".
The split comes months after 39-year-old Brand said he was "very happy" with Khan and that the relationship was "very different".
At the time, he said: "I'm going to be really kind in this relationship. I haven't been out there trying to nail everyone. I'm very, very happy in a relationship that feels very different from anything I've experience before.
"I feel like its grounded in friendship and a sort of love and it's all well cool and exciting and stuff, but it feels different for me."
Brand was previously married to singer Katy Perry. They revealed their engagement in December 2009 before tying the knot in October 2010. After 14 months of marriage, the comedian filed for divorce in December 2011, citing irreconcilable differences. Their divorce was finalised in July 2012.
Khan was formerly married to Imran Khan. The couple have two teenage sons together.
