Last month, the actress confirmed she and husband Will Kopelman were expecting their second child together.
On Wednesday night, the 38-year-old attended the Beauty Inc. Awards in New York and said she was expecting another girl.
Accepting the Best Newcomer Award for her cosmetics brand, FLOWER Beauty, the actress told the audience: "I am so proud to accept this in celebration of women, including my daughter and my future daughter."
A representative for Barrymore later confirmed the baby's gender to E! News.
At the time of announcing her pregnancy, the actress said she felt "really lucky" to be a parent for the second time.
Speaking to People magazine, she added: "When I found out I was overjoyed. It always feels so surreal, it's incredible."
Barrymore and art consultant Kopelman started dating in early 2011. They announced their engagement in January 2012, and married in June of that year. In September 2012, the couple welcomed their first child together, a daughter called Olive Barrymore Kopelman.
