The ceremony was held just days after President Francois Hollande signed the same-sex marriage bill, making it legal.
Amid tight security in Montpellier, Vincent Autin and Bruno Boileau said 'I do', in front of 600 guests, with more than 100 journalists also in attendance.
Wednesday's ceremony was broadcast live on French television, with the Socialist mayor of Montpellier, Helene Mandroux, conducting the ceremony. Extra police were drafted in to the town following fears that protests would be staged near the event.
France has seen violent protests break out following the new legislation, with many arguing the bill undermines an essential building block of society. On Sunday, around 150,000 people took to the streets of Paris in protest against the bill.
Mr Autin, 40, met Mr Boileau, 30, in 2006. Now the couple are married, they have expressed their interest in adopting a child.
France is the 14th country to legalise gay marriage after New Zealand passed a same-sex marriage bill last month.
