The couple's baby girl was born on Thursday morning, according to Digital Spy.
The television presenter posted a number of messages onto social networking site Twitter as he waited in the hospital.
At one stage before the birth, Humphrey's was waiting outside Kingston Hospital's radio station and he tweeted: "Tempted to go and do some presenting while waiting."
He then added: "I don't seem to be able to do or say anything without seeing myself as one of the blokes from One Born Every Minute."
He also posted a photo of his daughter's hand just moments after she was born.
In the tweet, which also revealed his child's name, Humphrey's wrote: "Thank you Florence Aurelia Alice Humphrey for a tender moment I'll never forget, and to Mum for making it possible x."
Humphrey's and his wife have been childhood sweethearts. The couple married in August 2007.
