One of the 600 guests, who attended the reception on Friday 29th April 201, is selling the cake in an internet auction.
The cake by Fiona Cairns, comes in the presentation tin commissioned and designed by Peter Windett and Sally Mangum. The printed compliments slip from TRH The Prince of Wales and The Duchess of Cornwall is included.
This is accompanied by the 16 page Order of Service from Westminster Abbey which includes the vows, hymns, prayers and Blessing from the Wedding.
The anonymous owner is expected to raise at least £1,000 for the cake.
Bristol-based PFC Auctions, are handling the sale.
Kylie Whitehead, of PFC Auctions, said: "It's estimated that two billion people watched the wedding on TV and this is the first piece of the wedding cake to appear at auction, so it's a very exciting lot.
"Slices of Diana's cake have been known to sell for more than £1,000 and we expect this slice will bring a similar amount.
"Together, the two artefacts make a superb, and potentially valuable, document of the day."
Bids can be submitted online from May 3.
